R287A- , D295A- , Y300F- , and R301A- mutants display increased Kd values that are 2 orders of magnitude higher than the wild type. The Y300A- mutant exhibits a 36-fold higher Kd value 58.4 M ; . The results confirm the reduced affinity of the E1b mutants for ThDP, as suggested by their increased Km values derived from E1b decarboxylation and the overall activity assays. E1b Mutations Impede Activity for Reductive Acylation-- Effects of mutations in the phosphorylation loop region on reductive acylation of lip-LBD from E2b Reaction 3 ; were deciphered. In the assay for reductive acylation, isolated recombinant lip-LBD instead of the natural lipoylated full-length E2b was used as substrate. The observation of mucosal abnormalities in Ilocano capillaria infected and noncapillaria infected subjects which led to the suggestion that there might exist certain anatomical and or functional peculiarities in these individuals which predispose them to the disease. The mechanism by which C. philippinensis causes malabsorption and protein-losing enteropathy has not been defined. It may be related to mucosal penetration and lymphathic obstruction, to the size of the worm burden, elaboration of toxins, interference with bile -salt function, changes in intestinal bacterial flora, or immunoglobulin deficiencies.
