Adrenoceptor Alkylation Studies with PBZ: Effects on Agonist-Induced Initial Constriction. To further determine whether the increased reactivity to adrenergic agonists seen in veins was due to differences in adrenergic receptor concentrations, we incubated the vessels with the -adrenoceptor alkylating agent PBZ 0.3, 10, and 30 nM ; and compared the effects on the NE- or PE-elicited initial peak constriction. Incubation of sham veins with PBZ 0.3 nM ; did not affect their initial constriction in response to NE Fig. 4A ; . However, higher PBZ concentrations 3, 10, and 30 nM ; produced a significant concentration-dependent reduction in the NE-elicited peak constriction Fig. 4A ; . DOCA-salt mesenteric veins were more resistant to PBZ-alkylating effects as the peak contractile response to NE was significantly inhibited only at the highest 30 nM ; PBZ concentration Fig. 4A ; . A 76-year-old man presented to the hospital ED at 10 complaining of severe orthopnea and a 1-week history of gradually worsening dyspnea and fatigue. The patient's spouse, who drove him to the hospital, also reported that she observed several days of intermittent confusion and poor appetite. The patient's history was notable for ischemic cardiomyopathy with an LV ejection fraction of 25%, coronary artery disease and previous myocardial infarction, and hypertension. Two weeks prior to presentation, the patient had a gout flair, which was treated with indomethacin 50 mg twice daily. Other outpatient medications included carvedilol 25 mg twice daily, losartan 50 mg daily, spironolactone 25 mg daily, digoxin, and furosemide 40 mg daily. Presenting BP was 92 64 mm Hg. The patient was dyspneic and in obvious distress. Jugular venous pressure JVP ; was to the angle of the jaw, and crackles were audible. He had a gallop rhythm and mitral regurgitation. The lower extremities were cold with 1 + to edema to the knees. The ECG showed left bundle branch block. Laboratory results are shown in Table 4. The ED physician's diagnosis was ADHF. The elevated BUN and serum creatinine concentrations indicated concomitant renal dysfunction. The patient was receiving optimal medication treatment, suggesting that decompensation was precipitated by the.

Only 21 of the 120 PBZ and GA air samples collected for glycols specifically ethylene, propylene, 1, 3 butylene, diethylene, and triethylene glycols ; during this investigation had detectable amounts of these substances. Excluding the results obtained from two GA air samples situated directly adjacent to the on-stage smoke machines used in LES MISERABLES, all of the remaining glycol concentrations were extremely low, ranging up to 2.1 milligrams per cubic meter mg m3 ; . These concentrations were well below the OSHA PEL for ethylene glycol of 127 mg m3 short-term exposure limit of 15 minutes ; . There is no NIOSH REL for ethylene glycol; however, because of the potential teratogenicity and the known respiratory irritation at the level chosen for the OSHA PEL, NIOSH has suggested that the current OSHA PEL be revised. There are no OSHA, NIOSH, or ACGIH exposure criteria for the other glycols. Air samples both PBZ and GA ; were collected for mineral oil mist during a dress rehearsal of MISS SAIGON. Concentrations ranged up to 1.35 mg m3, TWA over duration of the play. The highest levels were measured in GA samples positioned on stage. All of the measured concentrations were well below the OSHA, NIOSH, and ACGIH exposure limits of 5 mg m3 for up to a full-shift 8 to 10 hours ; TWA exposure. All 224 actors from nine Broadway productions completed questionnaires. Of this group, 134 questionnaires 60% ; were from actors appearing in the four productions using theatrical smokes, and 90 questionnaires 40% ; were from actors appearing in the five control productions. When compared to actors from the non-smoke productions, actors from two or more of the four productions utilizing theatrical smoke reported experiencing significantly greater prevalence of nasal symptoms sneezing, runny or stuffy nose ; , respiratory symptoms cough, wheeze, breathlessness, chest tightness ; , and mucous membrane symptoms sore throat, hoarseness, dry throat, itchy, burning eyes, dry eyes ; during their performances for the week prior to the survey. Although some of the constituents of theatrical smoke primarily the glycols ; have irritative properties, the reason for the high symptom prevalence in the productions that use theatrical smoke is not clear, since the time-weighted average concentrations of the glycols measured during the performances were quite low. It is possible however, that the smoke concentrations could be sufficiently high during the short periods of time that the smoke is generated to contribute to the symptoms reported by the actors.

We have reduced complexity over the past few years and renewed our product range. Low-margin products have been pruned to reduce the total number by 20%, while the business has been simplified by transferring some smaller customers to our distributors, where they can be better served. We are one of the industry leaders when it comes to introducing "greener" chemical products that are less toxic to the environment in applications such as offshore oilfield drilling and hard surface household cleaners. These products can be used in aqueous solutions or are more biodegradable. The European Union's REACH directive on product registration, which comes into effect in 2007, is another challenge for the industry. We have therefore been testing our products and have led various industry association consortia to use this regulation to our competitive advantage and pegfilgrastim.
In this particular study, using a combination of GCV treatment and quantitative PET, we provide proof-ofprinciple data for the use of the Sindbis viral vector system for GDEPT. The model tested is based on ES-2 Fluc cells derived from a clear cell ovarian carcinoma, known for its resistance to a variety of chemotherapeutic agents. Intraperitoneal inoculation of ES-2 Fluc cells in SCID mice induce an advanced disease characterized by rapid and widespread metastases within the peritoneal cavity. Our previous study indicated that the Sindbis vector, in the absence of any cytotoxic gene payload, is capable of systemic targeting and growth suppression of ES-2 Fluc tumor metastases 25 ; . In the present study, we also observed a similar therapeutic effect of Sindbis tk vector without the prodrug GCV Fig. 3A ; . The vector alone achieved significant tumor growth suppression P 5 0.0019, 2-way ANOVA ; , which is largely attributed to the apoptotic nature of the Sindbis vector. Coadministration of GCV with Sindbis tk treatments significantly enhances the therapeutic effectiveness of Sindbis tk by taking advantage of the expression of HSVtk in Sindbis tk-infected tumor cells Fig. 3B ; . Activated GCV not only enhances the killing of the infected tumor cells but also causes bystander effects by transfer to the surrounding untransfected tumor cells. Because the infected tumor cells are doomed to an apoptotic fate regardless of the GCV treatment, the enhancement of the therapeutic efficacy of the Sindbis tk vector is most likely due to the bystander effect, or a combination of both. In addition, the tumor apoptosis induced by Sindbis infection might enhance the bystander effects of GCV, as activated GCV could be transferred to surrounding tumor cells via apoptotic vesicles 28 ; in addition to gap junctions 19 ; . Beside a proper vector system, cancer GDEPT therapy would greatly benefit from a means to noninvasively monitor the GDEPT enzyme activity after vector treatments in vivo. Such capability could improve the Sindbis-based HSVtk GCV GDEPT in clinical settings by providing important information to address 2 critical questions: i ; Do the vectors systemically target tumor cells and spare normal tissues? ii ; Do the tumors have sufficient expression levels of the enzyme for tumor eradication by subsequent prodrug activation? In addition, monitoring during the therapy could facilitate optimizing the dose and dosing schedule of the prodrug to reduce unwanted side effects. Recent advances in optical and radionuclide imaging technology provide several methods for noninvasive monitoring of marker gene expression in living animals.
