When dyn is assembled around lipid templates, GTP hydrolysis induces a conformational change. Depending on the composition of the lipid, this conformational change can cause either constriction of the dyn spirals resulting in vesiculation Sweitzer and Hinshaw, 1998 ; or an increase in the distance between the rungs of the assembled dyn spiral Stowell et al., 1999 ; . Together these in vitro observations have led to a variety of working models proposing that dyn functions as a mechanochemical enzyme Hinshaw and Schmid, 1995; Warnock and Schmid, 1996; Smirnova et al., 1999; Stowell et al., 1999; Kozlov, 2001; for review, see Sever et al., 2000b ; . Although differing significantly in their details, these models all suggest that dyn's high, assembly-stimulated rate of GTP hydrolysis would be required for pinching off vesicles from the plasma membrane. Several laboratories have indeed established that overexpression of dyn GTPase domain mutants defective in GTP binding and or hydrolysis inhibits endocytosis in mammalian cells Herskovits et al., 1993; van der Bliek et al., 1993; Hill et al., 2001; Marks et al., 2001 ; . However, contradictory results were obtained with the use of dyn GTPase effector domain GED ; mutants. GED is an intramolecular GTPaseactivating protein GAP ; that is required for dyn's assemblystimulated GTPase activity but not for its basal rate of GTP binding and hydrolysis Sever et al., 1999 ; . Overexpression of these GAP-impaired dyn mutants accelerates the ratelimiting step in endocytosis leading to an increase in the rate of formation of constricted coated pits Sever et al., 1999, 2000a ; . Depending on the specific GED mutant, this can, in turn, lead to an increased rate of receptor-mediated endocytosis. Together, these data suggest a complex relationship between dyn's GTPase cycle, its regulation, and the execution of distinct stages in the formation of clathrin-coated vesicles. As a means to explore this complex relationship we sought to use the overexpression of dyn mutants defective in distinct steps of its GTPase cycle to accumulate sequential intermediates of clathrin-coated vesicle formation. The dyn mutants were designed in analogy to other members of the GTPase superfamily and were predicted to arrest dyn in its GDP-bound, GTP-bound, or nucleotide-unoccupied state. Biochemical characterization of these mutants revealed unexpected results relating to their GTPase properties in that neither of the mutations resulted in a GTPase predicted to be "locked" in either the activated or inactivated state. The formation of constricted coated pits was inhibited by the dyn GTPase domain mutants that we generated; however.

The newly approved agent will be marketed in the United States under the name MycamineTM micafungin sodium for injection ; . "This is an important milestone in Fujisawa's commitment to develop novel compounds that help patients with serious diseases, " said Hideo Fukumoto, Chairman and Chief Executive Officer, Fujisawa Healthcare, Inc. "Mycamine will allow us to expand our depth in the anti-fungal market and offer patients another treatment option to fight or prevent a life-threatening candida infection." Currently, Fujisawa markets the anti-fungal AmBisome a liposomal formulation of amphotericin B ; in the United States. According to a recent study, invasive candidiasis kills 10 - 40 percent of infected immunocompromised patients. A number of chemicals are known to produce developmental neurotoxic effects in humans and other species. For example lead and methyl mercury affect brain development but effects on fertility, the immune system, metabolism of foreign substances and development as a whole have been observed. 43. A recent proposal for an OECD guideline for "Developmental Neurotoxicity Study", draft TG 426 ; has been developed based upon an US EPA guideline. Developmental neurotoxicity studies are designed to develop data on the potential functional and morphological hazards to the nervous system arising in the offspring from exposure of the mother during pregnancy and lactation. These studies identify changes in behaviour due to effects on the central nervous system CNS ; and the peripheral nervous system PNS ; . As behaviour is affected by the function of other organs such as liver, kidneys and the endocrine system, toxic effects on these organs in offspring may also be reflected in general changes in behaviour. No single test is able to reflect the entire complex and intricate function of behaviour. For testing behaviour, therefore, a range of parameters, a "test battery", is used to identify changes in individual functions. Methodology employed in behavioral teratology is described in several reviews Adams 1986; Francis et al., 1990; IPCS 2001 ; . Catalyst wetting: The wetting efficiency can be defined as the averaged fraction of the external particle surface that is covered by liquid as shown in Fig. 1.9. It should be mentioned that in most cases the particle is internally completely wetted by capillary effects, unless in the case of very exothermic reactions, where the reaction heat release can volatilise the liquid trapped in the pores [167]. The incomplete wetting of catalyst particles in TBRs has two causes [156]. The first one is because of liquid maldistributions on reactor scale, that leave certain portions of the bed poorly irrigated. This problem can be readily addressed through a proper design of liquid distributors, as well as proper packing procedure. However, incomplete catalyst wetting is inherently related to TBRs at low liquid flow rate, because there is not enough liquid to cover all the catalyst surface. The catalyst wetting can be improved, not only by increasing the liquid flow rate, but also by increasing the gas flow rate or operating pressure, because both factors increase shear stress on the gas-liquid interface, which spreads the liquid on the catalyst surface [168]. Conversely, the operation at elevated temperatures results in a decrease of catalyst wetting [156] and minoxidil.
