A single-plaque isolate of polyoma virus obtained in these laboratories by Dr. I. Hellstrm was used. The original virus source was obtained from Dr. B. E. Eddy and designated 195611C. The history of this virus was described elsewhere 4 ; . One single pool of virus titrating 1.8 X IO8 plaque-forming units ml was used in all experiments. CharcotMarie-Tooth CMT ; disease is the most common form of inherited peripheral neuropathies. Clinical, neurophysiological and histopathological criteria are used to differentiate CMT into demyelinating CMT or CMT1, neuronal CMT or CMT2, and spinal CMT. CMT1 and CMT2 belong to the HMSN, while spinal CMT belongs to the distal HMN. Up to now at least 26 loci and 9 genes for different inherited neuropathies have been assigned and identified in the human genome De Jonghe et al., 2000b ; . Genetic linkage studies mapped four CMT2 loci i.e. CMT2A, CMT2B, CMT2D and CMT2E at chromosome 1p35-p36, 3q13-q22, 7p14 and 8p21 respectively. CMT2C is an as yet unmapped locus De Jonghe et al., 2000b ; . We reported a large Italian CMT2 family linked to the CMT2A locus on chromosome 1p35-p36. Based on previously reported genetic data and the recombination events in our Italian family we could refine the CMT2A region from 16 to 10 cM. The clinical phenotype is uniform and similar to that reported in other CMT2A families, confirming intra- and inter-familial uniformity of the CMT2A phenotype. This CMT2A phenotype is distinct from that observed in CMT2 families associated with mutations in peripheral myelin protein zero MPZ ; or connexin 32 Cx32 GJB1 ; . In these latter cases, nerve conduction velocities NCVs ; can initially be slightly reduced or nearly normal but in elderly patients NCVs may drop to 25 m The NCV range is much broader than in the Italian CMT2A family, and the lower limit is much lower. Subsequently a c.998A C transversion mutation resulting in a Gln333Pro in the first exon of the neurofilament light NEFL ; gene was found to show complete co-segregation with the disease phenotype. We screened 40 unrelated patients diagnosed as CMT2 or intermediate CMT for the presence of mutations in the NEFL gene. Via DHPLC denaturing high performance liquid chromatography ; we detected an heteroduplex pattern in one of the NEFL-PCR fragments in the proband of a Belgian CMT family. DHPLC analysis demonstrated the same heteroduplex pattern in all patients from the same family, but not in their unaffected relatives. We subsequently sequenced DNA samples of three patients and found a double missense mutation at positions 22 and 23 from CC to AG the first exon of NEFL c.22C A + 23C G ; . This mutation creates an amino-acid change from proline to arginine at codon 8 P8R ; . This mutation was absent in 80 normal controls suggesting that this sequence variation is not a rare polymorphism. Linkage analysis with the Pro8Arg mutation in family CMT-56 resulted in a two-point LOD score of 3.61 in the absence of recombinants. The NEFL Pro8Arg missense mutation most likely destabilizes the head domain of the NEFL protein. The patients in the Belgian family present with a classical, although rather severe CMT phenotype and ibandronate.

A total of 53 patients were enrolled in the study, 40 in the MGDF group and 13 in the placebo group. MGDF and placebo were administered to three patients each before chemotherapy. However. in these 20 sets eight PMS events were found for the seven markers scored. The three markers farthest from their centromeres msr, act, and ery-IM3a ; accounted f o r seven of those events. The eighth early PMS event occurred at nic-13, which is approximately 12 map units from its centromere. The remaining three loci, sr-la, pyr and ac-17, approximately 12, 9 and 0 map units from their respective cmtrorr eres, exhibited no early PMS in this sample. Although the correlations between frequency of PMS and the distance of the locus from its centromere are somewhat crude, such correlations cannot be ignored. They suggest that a large part of the mitotic segregation occurring in the progeny of triploid zygotes is due to mitotic recombination rather than to chroTABLE 10.

Objectives. To examine whether high-resolution 18F-2-fluoro-deoxy-D-glucose 18FDG ; - positron emission tomography PET ; could visualise deterioration of cervical spinal cord function associated with variable degree of compression and determine its potential usefulness during assessment of compressive myelopathy. Methods. 18FDG-PET was performed in 23 patients requiring decompressive surgery for myelopathy. The preoperative results of high-resolution 18FDG-PET were compared with the neurological scores and magnetic resonance imaging MRI ; findings. Results. The standardised uptake value SUV ; of 18FDG utilisation rate of the cervical cord before surgery correlated with the preoperative r 0.497, p 0.016 ; and postoperative neurological score r 0.595, p 0.003 ; , and with the rate of neurological improvement postoperatively r 0.538, p 0.008 ; . The 18FDG utilisation rate did not correlate with the high signal intensity on T2-weighted MR images. Conclusions. Our results indicate that high-resolution 18FDG-PET imaging provides useful qualitative and quantitative estimates of impaired metabolic activity of the compromised cervical cord that correlate closely with the severity of neurological dysfunction and iloprost.
