Labeled thienylalanine was incorporated into gramicidin S to an extent equivalent to about 5% of the phenylalanine residues, with B. brevtis ATCC 9999 Winnick and Winnick, 1961 ; . Other analogues, such as norleucine and pfluorophenylalanine, were less effectively utilized. Also, Katz 1960 ; found that the proline analogues, pipecolic acid and azetidine-2carboxylic acid, could replace proline in the actinomycin peptides, though not in the protein of Streptomyces antibioticus.
The lesser shifts shown by the other carbonyl carbons are due to their helical arrangement, placing them at somewhat larger distances from the cation. This situation is depicted in Fig. 6, which is an end-on view looking from the solution end of the channel, with the distances in A of the Trp carbonyl oxygens from the midpoint of the channel given in parentheses. The double circle represents the mean position of the helical backbone atoms, and the peptide carbonyl oxygens are shown librated toward the helix axis as previously depicted for the libration mechanism 12 ; . What Fig. 6 shows is that the Trp carbonyl oxygens surround the bare cation laterally, with room for water preceding and following the cation in the channel. An important point to be made is that the carbonyls of the channel are able to compete with water, even with the proximity of the membrane lipid, and to do so with binding constants ofthe order of 102 to 103 M-1. Therefore, it is not required that more water be in the channel to increase the dielectric constant; the carbonyls themselves, when they have the proper orientation due to the local peptide conformation, can provide a greater free energy of solvation than water. This is consistent with the realization that, while the dielectric constant of water is about 80, the dielectric constant of N-methylacetamide, a model for the peptide moiety, is about 180 37 ; . This is also consistent with the NMR-observed chemical shifts. Whereas the carbonyl carbon resonances shift downfield on complexation with the cation because of a loss of electron density surrounding the carbonyl carbon, the resonance of the 'Na nucleus on going from water into the channel shifts some 20 ppm upfield 26, 27 ; , indicating the greater electron density accessible to the cation in the channel as compared to aqueous solution. While the structure ofthe cation-containing channel within a lipid membrane has not been determined by diffraction methods, a crystal structure of gramicidin A at low resolution has been obtained for the K + and Cs' complexes 3 ; . The apparent length and diameters of the helical structure are within tenths of an A the channel structure described some 10 years ago ref. 5; ref. 12, table of I 3 helix parameters ; . In the crystal structure, the cation-binding sites are seen to be within the end-to-end aligned helices and to recur with separations of 21 A and 5 A, but it is not possible to say whether this indicates two sites straddling the midpoint of the channel and separated by 5 A whether it is two sites separated by 21 A.

Our conclusion, that the elementary conducting unit is a dimer, conflicts with that reached by Stark et al. 1986; see also Strassle et al., 1989 ; . Group A and B isolates cocirculated during most epidemics in Argentina and Chile, with the relative proportions varying from year to year. During 1996 and 2000 only group A strains were isolated in Argentina, although few strains were characterized in 1996 as to drive conclusions in this respect. In Chile, group B strains were detected by IF during 2002, but these strains did not grow well enough to perform a reliable ELISA. Different prevalences of RSV groups were observed in both countries throughout the period studied, as shown in Table I. The strain-specific variability of 172 viruses from group A was revealed with MAbs 021 5G, 021 and 68G Fig. 1 ; . All of them are directed against variable epitopes that have been mapped in the C-terminal third of the G glycoprotein, except 021 5G that maps in the N-terminal hypervariable region [Garcia-Barreno et al., 1989; Rueda et al., 1991, 1995; Martinez et al., 1997]. The antigenic variability within group A was represented in 23 different antigenic patterns Fig. 1 ; . Physical characteristics of pores and channels Gramicidin is highly polymorphic, being able to adopt a wide range of structures with dierent topologies, orientations and hands. The two most common forms of gramicidin are the double helix DH ; , a dimer in which two monomers are intertwined [Fig. 1 a ; ], 4 and the helical dimer HD ; , a dimer in which two helical monomers are joined end-to-end [Fig. 1 b ; ].5h7 Both the double helix and helical dimer forms have b-sheet-like hydrogen bonding patterns, diering in the number of intra- and inter-molecular hydrogen bonds, and the helical rise per residue. The HD form is commonly known as the "" channel , whereas the DH structures are often referred to as "" pores . In ion-free methanol solution, gramicidin has been shown to exist as four interconvertible double helical conformers, referred to as species 1, 2, 3 and 4, diering in helical sense and chain orientation.8 The components of this equilibrium mixture can be separated using chromatographic techniques, allowing characterisation of the individual species by circular dichroism CD ; spectroscopy. Veatch and co-workers8 proposed that species 1 and 2 are left-handed parallel DHs diering in the staggers between the ends of their chains, species 3 is a left-handed antiparallel DH, and species 4 is a right-handed parallel DH. The relative abundance of these four species in a given solution depends on the nature of the solvent.9 Inuences on the equilibrium between DH and HD forms The equilibrium between the DH and HD forms of gramicidin can be shifted by a number of physical conditions. The principle inuence appears to be the chemical nature of the environment. Whilst in organic solvents the DH forms are almost exclusively found with the relative proportions of the four DH species present determined by the nature of the solvent ; , 9 in most lipids the HD form predominates. However, if the membranes are prepared from lipids with polyunsaturated fatty acids, 10 this has been shown to bias the equilibrium towards the DH forms in small unilamellar vesicles SUVs ; , and conformational transitions between the DH and HD forms appear to occur faster in bilayers with polyunsaturated acyl chains.11 In addition, in SUVs the balance between DH and HD forms can be aected by the chirality of the lipid molecules12 used to produce the bilayers. It has also been shown that the relative proportions of DHs and HDs present depend on the solvent and temperature used during the preparation of the SUVs the so-called "" solvent-history dependence ; . The conformation which is initially adopted when gramicidin is incorporated into membranes is determined by the solvent in which it was dissolved during sample preparation.13h16 This is not necessarily the conformation that is nally adopted at thermodynamic equilibrium, however. Sychev et al.10 assert that while the initial conformational state of gramicidin in hydrated lipid bilayers is aected by the solvent, the equilibrium conformational state after heat incubation ; is determined by the lipid structure rather than the nature of the solvent. When taken altogether, these studies show that considerable variation in the relative proportions of the HD and DH forms can be achieved in SUVs. Quite a dierent picture is seen for gramicidin in bilayer lipid membranes BLMs ; used for conductance studies. Under ordinary conditions, essentially only HDs are detected in these and guarana.

A variety of studies were undertaken to confirm that the markedly diminished efflux of folic acid and MTX in PyrR100 cells was due entirely to a change in membrane transport and not due to: 1 ; increased anti ; folylpolyglutamylation, 2 ; increased compartmentation of folate in mitochondria, 3 ; sequestration by a highly overexpressed anti ; folatebinding protein, or 4 ; decreased plasma membrane potential. After loading cells with 1 M [3H]MTX for 1 h, cell extracts were analyzed by HPLC for MTX and its polyglutamate derivatives. PyrR100 and AA8 cells both contained 85% of the radiolabel as the MTX monoglutamate data not shown ; . The remaining 15% of radiolabel was associated with decreasing amounts of diglutamate, triglutamate, and tetraglutamate in both cell lines. Thus, the vast majority of radiolabeled MTX in both PyrR100 and AA8 cells remains in the monoglutamate form after 1 h incubation with [3H]MTX. HPLC analysis of cell extracts following 1 h incubation with 1 M [3H]folic acid in the presence of 5 M TMQ revealed that greater than 95% of the radiolabel in both PyrR100 and AA8 cells represented folic acid monoglutamate. Hence, there was no metabolism of folic acid under these conditions. To examine the levels of compartmentation of folic acid in mitochondria, cells were loaded with 1 M [3H]folic acid in the presence of 5 M TMQ for 1 h, following which mitochondria were isolated in isotonic or hypotonic buffer.
