Using IL-7-deficient DC, we examined if at least part of the DC-induced CD4 + T cell survival signal could be mediated by IL-7. Such DC were as efficient as wt DC promoting T cell survival Fig. 3B ; , providing a first indication that IL-7 is not essential for the DC-induced T cell survival signal in this system. In order to make the XML syntax amenable to a rigid formal description, RELAX NG defines a full syntax, a simple syntax, and precise conversion rules that govern conversion of schemas from the full into the simple syntax. While the full syntax has a richer set of language elements making it more convenient to use ; , the simple syntax is very restricted in its language constructs, but rather cumbersome and lengthy. For instance, the full syntax provides means for including external content, whereas simplification requires all references to be resolved, and all external resources to be inserted in-place. To determine the amount of metabolite in the serum, the following extraction procedure was employed: to 2.5 ml of serum, 0.1 ml of n-propanol was added and the mixture was heated at 60C for 10 min. The mixture was acidified to pH 2 and extracted three times with 6-ml portions of n-hexane-i-butanQl vol vol, 3: 2 ; . The combined organic layers were evaporated to dryness, and the residue was dissolved in the appropriate volume of ethylacetate. From the respective samples 101I was spotted onto the plate. Four standards 10 Al of four dilutions of the analytical grade penicillin in 0.2 M NaCl ; were chromatographed simultaneously on each plate. Prior to chromatography, the spots originating from aqueous samples were subjected to spot chromatography by applying 5 , ul of acetone at their centers. Thus, tailing because of overloading of the plate was avoided. The chromatogram was developed at room temperature with a mixture of 0.3 M NaCl in 0.05 M potassium phosphate buffer pH 5.6 ; acetone vol vol, 2: 1 ; . After chromatography the substances were located by fluorescence quenching at 254 nm. The parent compound, the metabolite, and the standards were recovered by scraping the spots from the plate and extracting with 0.4 to 0.5 ml of water saturated with methylethylketone. After centrifugation the amount of penicillin in the supernatant was estimated by means of the agar cup diffusion technique, using a susceptible microorganism commonly S. lutea ATCC 9341 ; as the test organism. From the standards, reference curves were made. The recovery from the silica gel by this procedure was about 85 to 90% for dicloxacillin and flucloxacillin and about 90 to 95% for cloxacillin and oxacillin. The recovery as established from the simultaneously chromatographed references was not significantly different from 100%. The lowest amount of penicillin that could be determined accurately in this way was 0.3 jug for oxacillin, 0.6 ug for dicloxacillin, and 1 , ug for cloxacillin and flucloxacillin. The precision of the procedure was 10 4%, as estimated from eight different samples; each sample was estimated four times. The method implies that the metabolites are estimated in equivalents of parent compound activity. Therefore, it was preferable to express the amount of metabolite as the percentage of the estimated total amount of penicillin in the respective biological fluid.
