The pale-staining variety is often larger than the darkstaining variety. Occasionally these granulocytes may be small with a glassy appearance and with an indistinct nucleus. However, they usually appear swollen and variable in shape. The nucleus is typically multilobulated or divided into four separated nuclei; it stains a very light blue. The cytoplasm of such cells contains granules, which show brownian movement if the specific gravity of the urine is not too low. As this brownian movement causes a constant variation of reflected light, the pale-staining white blood cells are often termed "glitter cells." Previously these cells were regarded as specific for pyelonephritis. However, it has been shown that glitter cells can be seen in almost any active urinary tract infection. Apparently pale-staining white blood cells are still living and are unable to bind dye molecules. b. Dark-Staining Variety. Dark-staining white blood cells represent inert forms that have undergone autolysis and thus have binding sites available to take up the dye. These cells have dense, purple nuclei. Granules in the cytoplasm are either not evident or are characterized by a purple granularity. They are generally uniform in size and occur commonly in lower urinary tract infections with renal involvement. 3-23. EPITHELIAL CELLS a. Renal Epithelial Cells. Renal epithelial cells are only slightly larger than white blood cells. They have a very thin rim of cytoplasm and a round nucleus with a dark band of chromatin at the periphery. The cytoplasm stains an orange-purple color. b. Bladder Epithelial Cells Caudate Cells ; . Bladder epithelial cells are frequently boat-shaped navicular cells ; , and some appear to have tails. The cells with tails are often called caudate cells. These cells also have a round nucleus. However, they have more cytoplasm than the renal cell, and the cytoplasm is distinctively pale blue with occasional inclusions. c. Squamous Epithelial Cells. Squamous epithelial cells have small, dark purple, pyknotic thickened, shrunken ; nuclei, and extensive pale purple cytoplasm. They frequently occur in sheets. It is not possible to differentiate squamous cells by their site of origin.

Nonsteroidal anti-inflammatory drugs, which inhibit cyclooxygenase COX ; activity, are powerful antineoplastic agents that exert their antiproliferative and proapoptotic effects on cancer cells by COX-dependent and or COX-independent pathways. Celecoxib, a COX-2specific inhibitor, has been shown to reduce the number of adenomatous colorectal polyps in patients with familial adenomatous polyposis. Here, we show that celecoxib induces apoptosis in the colon cancer cell line HT-29 by inhibiting the 3-phosphoinositide-dependent kinase 1 PDK1 ; activity. This effect was correlated with inhibition of the phosphorylation of the PDK1 downstream substrate Akt protein kinase B PKB ; on two regulatory sites, Thr308 and Ser473. However, expression of a constitutive active form of Akt PKB myristoylated PKB ; has a low protective effect toward celecoxib-induced cell death. In contrast, overexpression of constitutive active mutant of PDK1 PDK1A280V ; was as potent as the pancaspase inhibitor, to impair celecoxib-induced apoptosis. By contrast, cells expressing a kinase-defective mutant of PDK1 PDK1K114G ; remained sensitive to celecoxib. Furthermore, in vitro measurement reveals that celecoxib was a potential inhibitor of PDK1 activity with an IC50 3.5 M. These data indicate that inhibition of PDK1 signaling is involved in the proapoptotic effect of celecoxib in HT-29 cells and arthrotec.
The proposed developmental scheme described here was based on examination of numerous sections of infected nuclei, many of which, contained all the pu.tative stages of development. Presumably complete capsids had squared ends and were of moderate density. The specialized apex, which was at the open end of the envelope, was considerably denser than the remainder of the capsid and sometimes apically serrate or frayed Fig. 5 ; . Often more than one viral particle was associated with a single piece of membrane and these particles were usually at different stages of development Figs. 4 and 6 ; . The rare occurrence of 2 viral particles apparently within a single envelope probably resulted from such associations Fig. 7 ; . Empty capsids surrounded by an open-ended tubular envelope and assumed to be complete were usually 65 to 7 210 to 280 nm. However, there were capsids as short as 95 nm and as long as 760 nm that appeared to be complete because they were either entirely enveloped or the envelope formed a tube that extended the entire length of the capsid Fig. 8 ; . Only one capsid and one nucleocapsid that appeared to completely lack envelopes were detected in the available material. It is proposed that the nucleoprotein o the core was f inserted centrally along the length of the capsid, first forming a helical filament that extended the full length of the capsid Fig. 9 A ; , and that the filament lost its helical appearance as it broadened during further filling of the capsid with nucleoprotein Fig. 9B, C ; .With a diameter of 95 to 110 nm, completed nucleocapsids were considerably broader than empty capsids Fig. 6 ; . They were 190 nm to 540 nm long, although most were between 235 and 280 nm long Figs. 5 and 9 ; . A newly completed nucleocapsid was straight, with the envelope fitting loosely along its length. The apex at the closed end of the envelope was rounded and the squared specialized apex at the open end was like that of the empty capsid Fig. 6 ; . The capsid was l l to thick along the length of the core, and sometimes separated slightly from it. The capsid was less electron dense than the core, and easily distinguished from it and avalide.
